Inflammatory bowel diseases (IBDs) are thought to result from an interplay of lifestyle and environmental factors together with predisposing genetic susceptibility leading to abnormal immune reactions against intestinal microbial antigens. IBDs develop progressively, i.e. microbial changes can be detected years before the onset of IBD with relatives of IBD patients being at particular risk for developing the disease. The aim of this project is to identify molecular and immunological signatures that predict the clinical onset of IBD. We will use novel technologies to characterize the T cell response against microbial antigens in high-risk relatives and IBD patients and to investigate the link between microbial dysbiosis and altered immune reactions.The successful candidate will work with state-of-the-art technologies to detect, isolate and characterize antigen-specific T cells against defined candidate microbes by multi-dimensional flow-cytometry, as well as single cell and next generation sequencing technologies. The identification of molecular and immunological signatures that predict the clinical onset or course of IBD may provide the basis for early and targeted therapeutic intervention.
